Five lives have been claimed and 720 adverse drug reactions have occurred in Britain since the launch of France’s Sanofi-Aventis SA’s weight-loss drug, Acomplia, two years ago; this, according to a posting on the United Kingdom’s drug regulator’s Website this week. Acomplia—known generically as rimonabant—had been slated to be a multi-billion dollar seller; however, following its rejection in the United States by a panel of US Food and Drug Administration (FDA) experts a year ago, its success has faltered. The team of US experts claimed the drug increased the number of psychiatric events, such as depression and suicidal ideation. Acomplia posted sales of 79 million Euros in 2007.

Acomplia is a new type of anti-obesity drug that blocks certain brain receptors that regulate appetite. In a 2006 study, Acomplia emerged as a promising diabetes treatment when patients who took Acomplia for a year reported improved blood sugar and cholesterol control in conjunction with moderate weight loss. A more recent study of Acomplia in obese heart patients revealed that nearly half—40 percent—of those patients studied developed psychiatric problems. Another study published last month raised concerns about using drugs such as Acomplia in children.

Despite this, Acomplia remains on shelves in the European Union with strengthened safety warnings and cannot be prescribed to patients suffering from depression. According to the Medicines and Healthcare Products Regulatory Agency’s (MHRA) website there has been one case of suicide in the period up to May 9, 2008, and two suicide attempts. A Sanofi spokeswoman said most of the five reported fatalities occurred before 2008 and that the report referred to the aggregated total of adverse reactions recorded in reports received by the MHRA since 2006. The other four deaths related to two cases of fatal heart attack, one sudden death, and one case of infectious disease.

Meanwhile, Merck &Company in the US is developing a similar drug—taranabant—which is in late-stage development and is also showing promise of enabling weight loss; however, studies have linked taranabant to increased psychiatric side effects such as those exhibited in Acomplia.

Merck has been in the news most recently after agreeing to pay $58 million to Massachusetts, 28 other states, and the District of Columbia, to settle lawsuits that claim Merck allegedly used deceptive marketing to promote its popular painkiller Vioxx.

Vioxx is in a class of drugs called nonsteroidal anti-inflammatory drugs (NSAIDs) and works by reducing substances that cause inflammation, pain, and fever. A three-year study aimed at showing that Vioxx—at a 25-milligram dose—prevents recurrence of polyps in the colon and rectum was ceased when Merck discovered a higher heart risk compared to patients taking placebos. Prior to the Vioxx withdrawal, the FDA announced that patients taking Vioxx had a 50 percent greater chance of heart attack and sudden cardiac death and patients taking the highest recommended daily dosage of Vioxx had three times the risk of heart attack and sudden cardiac death as those not taking standard painkillers. Merck pulled Vioxx from the market on September 30, 2004 after acknowledging that Vioxx could significantly increase the risk of heart attacks.

Sanofi-aventis publishes positive judged results of his study Athena on the Multaq, his treatment of the fibrillation little finger, a disturbance of the cardiac rhythm being able to provoke vascular cerebral accidents.

The study presented to the Corporation of American Cardiac Rythmologie was realized in blind double, on 4,628 recruited patients in 37 countries.

His vocation was to evaluate a treatment intend for the taken one in charge of the fibrillation little finger in complement of the conventional treatments while measuring the impact on the reduction of the hospitalizations cardiovasculaires or the mortality.

She revealed that the Multaq, the commercial name of the molecule Dronédarone of Sanofi, diminished of 24% the risk of hospitalization of origin cardiovasculaire or of death in comparison with placebo.

The study also proved as the Multaq allowed a reduction of 30% of the risk of death cardiovasculaire in comparison with placebo (neuter substance). In addition, the treatment drove to a decrease of 45% of the risk of death for causes cardiac arrhythmia.

The undesirable effects the most frequently retrieved in comparison with placebo are type liqueur (26% against 21%), skin (10% against 8%) as well as an elevation of the sanguine créatine (4.7% against 1%), specified the n°4 world-wide one of the pharmacy.

TOWARDS A MARKETING BEGINNING 2009 TO THE WORE

“These results never were obtained for 20 years in this pathology”, underlined John Philippe Santoni, the director of the international development of sanofi-aventis at the time of a press conference.

“The study results are positive”, comments on for its part an analyst under cover with anonymity.

On the basis of these new data clinics, Sanofi foresees to submit of here the third quarter 2008 a request of authorization of placement on the market with the European Agency of the medicine as well as a probate request with sanitary American authorities.

John rock Lehner, director of the medical matters of sanofi-aventis, considered that Multaq could be marketed to the first quarter 2009 to the United States, for the file will be able to be examined in the six following months his deposit.

In Europe, it followed, the treatment could be on the market towards the end of next year.

Sanofi-aventis did not give indication on the potential sales of this medicine but underlined that the fibrillation little finger touches about 2.5 millions of persons to the United States and 4.5 million in the European Union.

John rock Lehner specified that the fibrillation little finger represented in terms of cares an annual cost of 3,000 Euros by patient.

In September 2006, sanofi-aventis had withdrawn his request of authorization of placement on the market in Europe of the Multaq, a week after wiping a file refusal from the

FDA.

The positive results of the study Athena could counterbalance the negative impression left by the launch project in Europe of a generic one of Plavix, the anticoagulant star of Sanofi, and by the invalidation to the United States of the patent of the ANTI-l’anti-thrombotique anti anti l’anti-thrombotique Lovenox.

They could allow the rebond of the title Sanofi that lost close to 23% since the year beginning. Second French stock market capitalization there are again eighteen months, Sanofi is today in fifth position, behind Total, EDF, ArcelorMittal and BNP Paribas.

Thursday, the action closed on a gain of 0.10% to 48.50 Euros while the CAC 40 was stable and that the European sectoral index progressed of 1.18%.

Acomplia (generic name rimonabant) has been an authorised medication in the EU since June 2006 for use in adult patients where the BMI exceeds 30 or the BMI exceeds 27 with associated risk factors such as diabetes. In all cases it can only be prescribed in conjunction with a hypocaloric diet and physical exercise.

In March the French government announced that acomplia was the first selective cannabinoid-1 receptor antagonist to be reimbursable by Social Security. Patients with prescriptions are entitled to recover 35% of the cost. The fact that no other medication has been approved for the subsidised treatment of obesity, gives acomplia a big market advantage in France.

In April, the Swiss government followed suit, placing acomplia on the mandatory reimbursement list both for the treatment of patients with type 2 diabetes with a BMI of 28 or more, and for the treatment of any patient with a BMI greater than 35.

But acceptance was not limited to Europe. April also saw an announcement from the Brazilian government that acomplia can be marketed to patients with a BMI over 30 or greater than 27 where there are risk factors present.

In July, the European Medicines Agency (EMEA) modified its blanket approval for acomplia. It formally accepted evidence that there may be psychiatric side effects and so issued a directive that acomplia should not be prescribed to patients who have been diagnosed with major depression or who are already receiving antidepressants. In one sense, there was nothing new about this because the risk was identified when approval was first given in June 2006. But the EMEA’s Committee for Medicinal Products for Human Use (CHMP) changed the level of warning.

The CHMP holds a general brief for monitoring the safety of medicines licensed for use in the EU. In June, it therefore requested Sanofi-Aventis, the French manufacturer, to submit all the available data it had in its possession on these side effects. In July, it concluded that the benefits of using acomplia continued to outweigh the possible disadvantages except in the cases where a patient was more seriously depressed. Thus, the warning now directs doctors either not to prescribe acomplia where a patient has been diagnosed as severely depressed, or to discontinue the use of acomplia if a patient subsequently becomes depressed. By publicising its findings, the CHMP was also taking steps to alert patients and their carers of the potential risk of using acomplia in relation to depression.

In November, the European Commission accepted EMEA’s positive impression of acomplia, reacting favourably to the results of the SERENADE study (Study Evaluating Rimonabant Efficacy in Drug-NAive DiabEtic Patients), in which newly diagnosed type 2 diabetic patients showed significantly improved glycemic control and a reduction in cardio-metabolic risk factors.

It is important to recognise that SERENADE is part of a more extensive Phase IIIb clinical trial program being carried out round the world. At present, there are more than 25,000 participants in eight further studies to study the effect of acomplia in the treatment of type 2 diabetes. These results are expected during the next two years.

Symptoms of adiposity: adiposity of the moderate degree - a short wind at movements, slackness of digestion; adiposity of an extreme degree - a short wind at the slightest movement, slackness of digestion, frustration of heart.

In a basis of adiposity infringement of a metabolism when processes of formation of fat of nutrients prevail of processes of its disintegration lays. Adiposity (fatness) almost always causes frustration of activity of an organism: work of heart is at a loss, blood circulation is broken, there is a short wind, fast fatigue, stability to infections decreases. To get rid of excess weight, it is necessary to consult, first of all, with the doctor.

Treatment of adiposity:

Methods of treatment of adiposity are subdivided into three basic groups.
1) not Medicinal methods: a rational low-calorie feed and increase of physical activity.
2) Medicinal methods: purpose of the preparations overwhelming appetite and reducing mastering of nutrients in an organism, and also the preparations normalizing a hormonal background.
3) Surgical methods of treatment of adiposity:

liposaction - removal of surpluses of fatty fabrics from under an integument;

Roundabout enteroanastomosys - the operation which is “switching off” from digestion of a part of a thin gut;

Gastric Band which are imposed on a stomach, reducing its volume and accordingly accelerating time of saturation;

abdominoplastics - elimination of the drooped and-or big stomach.

The appetite suppressing receptors are called cannabinoid receptors. They control the plasticity and also regulate signaling among neurons in the visual cortex.

The researchers looked at the drug rimonabant (Acomplia). Findings suggest obesity drugs that block the cannabinoid receptors severely disrupted the cortical plasticity in young mice, so researchers believe they may have the same effect on human children.

As a result, the scientists are urging caution in using these drugs on overweight children.

Sanofi-Aventis, the company that makes Acomplia, is awaiting FDA approval for its use in the United States. Other pharmaceutical companies have similar drugs in development.

If you have been exercising and eating a healthy diet but still cannot seem to meet your weight loss goals you might want to consider taking a weight loss or diet pill in addition to your regular routine to help you burn those extra calories each day that might be the big difference between burning fat or maintaining your current weight.

Some people have doubts as to whether weight loss pills work and whether they should try using one as an aid. Here are a few things you should ask yourself before determining whether or not they are right for you.

The first thing you need to ask yourself is whether you have really tried to lose weight by the standard methods of exercise and proper diet. If you are overweight, you can’t simply take a diet pill but continue to live the exact same lifestyle and expect to lose weight. You still need to make sure you are burning the same or more calories that you consume each day either by cutting out calories from your diet and meals or by exercising more to lose weight.

There are however, some pills that can help reduce the appetite and minimize over eating. The problem here is that if you cut back too much on calories, your body will burn less calories and if you stop taking the pills, you will begin over-eating again and put back on all the weight you initially lost as well as having a less aggressive metabolism. Optimal use of any diet pill is by following a proper diet with at least some exercise each day, although for some people, healthier eating and taking an additional diet pill is all they need to start losing those unwanted pounds.

By reducing your appetite just enough, some pills can help stop some people from over eating. This can be the difference between getting fatter or staying the same, which you can then slowly work on with some exercise or changes to your diet.

You can also research what ingredients are in the pills and see if there are any common foods or beverages that contain them and start adding them to your current diet.

Another thing you need to consider is cost. You might want to try a bottle first to see if you get any result but you really need to consider how much it will cost to use them for several months or even a year depending on how much weight you want to lose and whether you can afford to keep using them for a prolonged period of time. It is always advised that you consult with your doctor before undertaking the use of any diet pills as they might interfere with certain medications you may be taking at the same time.

The secret to weight loss through low fat cooking isn’t to cut all of the foods you love from your diet. It’s learning to make the foods you love in healthier ways.

If you want to lose weight, you have to burn off more fat than you consume. So cutting the amount of fat you have in your diet is the first step. And one way to do this is to substitute some of the high fat ingredients you cook with, instead using fat free or low fat alternatives.

You Can Eat Meat

A low fat diet doesn’t mean you have to give up meat. What is important is the type of meat you choose.

Removing the skin from chicken also removes a lot of the fat. With beef or pork, choose thin cuts. And if there is any visible fat on a cut of meat, trim it off.

Even on a low fat diet, you can still eat bacon and sausage. Sausages made with chicken or turkey contains less fat than beef or pork sausages. And turkey bacon is better for you than bacon make from pork. And it tastes good, too.

Cooking Your Meat

How you cook your meat also makes a difference. Chicken that has been baked, broiled or grilled is going to be lower in fat than chicken that’s been deep fried. Coating chicken with spiced breadcrumbs and then frying them can make a tasty substitute for deep fried chicken.

If you need to sauté your meat for one of your recipes, do so in a skillet that you have lightly sprayed with nonstick cooking spray.

Butter and Oil

nstead of using butter or oil to cook your eggs in the morning, spray the skillet with nonstick cooking spray instead. If you must use cooking oil, choose olive oil or canola oil over something like regular vegetable oil.

There is no denying that butter adds flavor to your dishes. But you can add flavor in other ways. For example, with egg dishes, you can experiment with all kinds of herbs and spices to give them more flavor. And, when you cook rice in fat free chicken broth, you taste buds won’t even miss the butter, and it tastes delicious!

Milk and Cream

A lot of the dishes you make that call for milk will work just as well with fat free milk as they would with whole milk. Dishes that call for heavy cream, you can substitute fat free evaporated milk.

And you can still make those delicious cheese cakes. Just substitute reduced fat cheeses for the ones you usually use.

Weight loss through low fat cooking doesn’t mean completely giving up on the foods you love. In most cases, it just means replacing some of the ingredients with healthier alternatives.

Acomplia is an oral prescription medication which has created waves in the weight loss market ever since its launch in the UK. Subsequent to which it has been launched in various other countries such as France, Germany, Ireland, Austria, Argentina, Denmark, Holland and Sweden. Some of the prospective countries which can witness the launch of this weight loss medication include Italy, Spain, and Mexico.

Use of Acomplia may cause a few adverse effects as a consequence of bodily changes in response to this medication. Yet, regardless of the adverse effects it sources such as anxiety and feeling of depression, Acomplia is still preferred by obese and overweight individuals who desire to lose weight.

“Prescription medication attaches the pre-requisite to procure a medical advice, but Alli can ease you of the hassle to run around doctor’s clinics. This is one reason why dieters round the globe are eagerly awaiting its launch which is likely to be around the month of July”.

Alli diet pill is presently under its pre-launch test. Glaxo-SmithKline, the drug-maker intends to provide 3-month supply to about 400 dieters prior to its launch in the stores.

This weight loss medication works by obstructing the fat absorption into the body. Consumption of this diet drug thereby is effective in giving ample opportunity to the already stored in fat to generate the required energy, assisting in trimming down of excessive fat. This is in turn consequential in effective weight loss.

From crash diets to killer workouts, it’s little secret that the weight loss regimens of the stars are not always healthy ones.

But a relative newcomer to the celebrity weight loss buzz — the attention deficit hyperactivity disorder (ADHD) drug Adderall — has a number of nutrition experts worried that those hoping to emulate their favorite celebrities could be putting their health at grave risk.

According to a report in the New York Daily News on Monday, a number of female celebrities have used prescription Adderall, even if they have not been diagnosed with ADHD. And reports also suggest that some of these starlets have resorted to crushing and snorting these pills as a way to deliver a quicker effect.

“Often, if we’re just beginning to hear of stories about inappropriate use of drugs, like Adderall, for weight loss, it means it’s already a significant problem,” says Jackie Newgent, a registered dietitian and New York City-based nutrition consultant.

Connie Diekman, president of the American Dietetic Association and director of university nutrition at Washington University in St. Louis, says that “few of my clients report using Adderall for weight loss, but I am very aware of the existence of this problem.

“While these identified risks are significant, the risk that is of biggest concern is the implication that, because celebrities do this, it must be safe,” Diekman adds. “Celebrities are viewed as role models — or at least idols — for many people, and … such potentially harmful behavior puts them and others [at] unknown risk.”

And as the number of legitimate prescriptions for the drug grows, some worry that the rates of its abuse could climb as well.

“I find the use of Adderall for weight loss particularly troubling,” says June Stevens, chair of the department of nutrition at the University of North Carolina at Chapel Hill. “So many of our youth take Adderall for attention deficit disorder. I fear it may lead to eating disorders and dependency on the drug as a weight loss aid.”

Sometimes you don’t have to look very hard for an investment idea. Pardon the pun, but obesity is a pretty big problem in the U.S. You certainly don’t need to work at Weight Watchers or a gym to see that Americans are overweight.

While gyms and diets are certainly one way to fix the problem, they don’t seem to be doing a whole lot of good; Americans have become more overweight with each passing year. Ultimately, Americans might get out of this mess the same way we got fat in the first place — a lazy fix, in the form of a drug.

Unfilled market
You’d think that weight-loss drugs would create a multibillion-dollar market, but that hasn’t happened yet.

Part of the problem is that the drugs just don’t work terribly well. In clinical trials supporting Abbott Laboratories’ (NYSE: ABT) Meridia, for instance, obese patients lost just 10-15 pounds more than patients not taking the drug. While every little pound helps, drugs like Meridia don’t seem to be the magic bullet that people are looking for.

The other problem is that the current drugs have side effects that drive prospective patients away. For example, GlaxoSmithKline’s (NYSE: GSK) Alli can cause gas and other unpleasant bowel changes. And while you might think that’d provide extra incentive to people taking the drug to stay away from fatty foods, those consequences are more than some people are willing to take.

In the works
The FDA’s refusal to approve Sanofi-Aventis’ Acomplia for marketing in the U.S. broke the field wide open. The first company to gain approval could open up the market, although me-too drugs that work better or have better side-effect profiles could easily overcome the first mover’s initial advantage.

Merck’s (NYSE: MRK) Taranabant and Pfizer’s (NYSE: PFE) CP-945598 both work by blocking the cannabinoid receptors in the brain. While stimulating the receptors (sometimes by smoking marijuana) gives you the munchies, blocking them causes you to lose your desire to eat. But like Acomplia, which also blocks the same receptor, the drugs can cause side effects like depression and even suicidal thoughts. Investors will get a closer look at Merck’s phase 3 data later this month, but early indications suggest that the drug isn’t as promising as Merck had hoped.

Arena (Nasdaq: ARNA) is targeting a different receptor, but it too could potentially have issues with side effects. Its phase 3 drug candidate, Lorcaserin, targets the same receptor as American Home Products’ (now Wyeth (NYSE: WYE)) fen-phen. One-year safety data from its two-year trial is due out this month, which should give insight into whether the drug causes the same heart-valve issues fen-phen did.

Farther behind in the pack, Amylin Pharmaceuticals (Nasdaq: AMLN) is testing a combination of its diabetes drug Symlin and an analog of leptin, a signaling molecule that regulates energy metabolism and body weight. The combination showed promising results in its phase 2 trial, but we’ll need a longer phase 3 trial to know for sure whether the drug combination really works. Like Novo Nordisk’s liraglutide, Amylin’s only downfall might be that its drug is injected, which could potentially scare off some patients if it doesn’t produce stellar results.

Risk vs. reward
The biggest problem for obesity-drug makers might not be the amount of pounds the drugs shed, but their safety in doing so.

All drugs have some side effects, but the acceptable level of side effects is generally tied to how likely one is to die of the disease. Thus, anti-cancer drugs can gain FDA approval with horrible side effects — think hair loss and nausea — but drugs that treat less severe diseases aren’t as liberally approved.

Given the history of side effects for weight-reducing drugs, investors should be cautious about the value they ascribe to drugs in the pipeline until all the long-term safety data is in. Waiting to invest might not fatten up your wallet as much, but it’ll sure keep you from losing your shirt — a side effect of failed phase 3 trials.
